Neuroblastoma is an extracranial malignant solid tumor originating from neural crest stem cells, exhibiting clinical behaviors from spontaneous regression to extensive metastasis. The lack of promising treatments highlights the need for novel research into its molecular foundations. The high proliferation rate of malignant neuroblastoma cells necessitates efficient energy metabolism; therefore, this study focuses on energy pathways and their role in tumor development. Genetic changes in neuroblastoma are associated with decreased TCA cycle enzyme activity and increased glycolysis. High-risk cases often harbor genetic alterations such as MYCN amplification, ALK mutations, or germline mutations in SDHA and SDHB genes. Mutations in SDHA and SDHB basically shut down succinate dehydrogenase activity, which forces a metabolic shift from normal mitochondrial energy production over to cytosolic glycolysis. In this project, α-TOS is used as an SDH targeted inhibitor to try and fix that mitochondrial dysfunction. Furthermore, high-risk patients typically exhibit elevated MYCN expression, which produces the N-Myc protein that inhibits the function of apoptosis-related genes. Epigenetic intervention via HDAC inhibitors offers a promising alternative strategy. SAHA has demonstrated the ability to reactivate the apoptotic pathways necessary for programmed cell death. Consequently, the synergistic potential of combining mitochondrial-targeted α-TOS with the epigenetic modulator SAHA is under investigation in neuroblastoma cells to evaluate its efficacy as a dual-action therapeutic strategy.
